Silencing of LncRNA steroid receptor RNA activator attenuates polycystic ovary syndrome in mice Scopus

© 2018 Elsevier B.V. and Société Française de Biochimie et Biologie Moléculaire (SFBBM) Background: Polycystic ovary syndrome (PCOS) is the most common endocrine disorder in women of reproductive age and has a prevalence of 1 in 15 women worldwide. This study aims to investigate the role of lncRNA SRA in the pathological processes of polycystic ovary syndrome (PCOS). Methods: Twenty five-day old female C57BL/6 mice received subcutaneous injection of 60 mg/kg dehydroepiandrosterone for 20 days to induce PCOS. Lentivirus containing lncRNA SRA-specific shRNA was subcapsularly injected into the ovaries of PCOS mice. Granulosa cell was primary cultured to explore the mechanism of DHEA-induced inflammatory responses. H&E staining was used to examine the histological changes of ovaries. ELISA was used to assess serum insulin level and proinflammatory cytokines and angiogenetic factors contents in ovary tissue. The expression levels of LncRNA SRA and proteins involved in the NF-κB signaling pathway were detected through Quantitative real-time PCR and Western blot. The nuclear translocation of NF-κB was observed by immunofluorescence and the activity of NF-κB-DNA binding was detected using EMSA. Results: Silencing of lncRNA SRA changed insulin release, attenuated ovary injury and reduced the production of angiogenetic factors in the PCOS mice. In addition, shRNA targeting lncRNA SRA inhibited DHEA-induced pro-inflammatory cytokines production and NF-κB nuclear translocation in the ovary of PCOS mice and primary granulosa cells. Conclusion: Silencing of lncRNA Steroid Receptor RNA Activator (SRA) attenuates polycystic ovary syndrome (PCOS) in mice. LncRNA SRA plays important roles in the development of PCOS.

Effect of calcium modification on solidification, heat treatment microstructure and toughness of high boron high speed steel Scopus

In this study, the as-cast and heat treated microstructures, hardness and impact toughness of high boron high speed steel with Fe-2.0 wt% B-0.4 wt% C-6.0 wt% Cr-4.0 wt% Mo-2.0% Al-1.0 wt% Si-1.0 wt% V-0.5 wt% Mn-x wt% Ca (x = 0.00, 0.08, 0.16, 0.24) is systematically investigated. The experimental results reveal that the as-cast microstructure of high boron high speed steel consists of α-Fe as matrix and eutectic borocarbide M2(B, C) (M=Fe, Cr, Mo, V, Mn). Borocarbide presents a continuous and reticular structure in alloy without calcium modification. After calcium modified, hardness of matrix increases because of solid solution strengthening. Besides, the continuous network of borocarbide is destroyed and possesses an isolated, fine and spherical structure. Calcium in high boron high speed steel has strong surface activity, which exists in the form of solid solution and mainly segregates at the interface between matrix and borocarbide. As a significant discovery, to calcium modified high boron high speed steel, the heat treatment can ulteriorly improve the shape and size of borocarbide. Moreover, the impact toughness of high boron high speed steel is remarkably improved by calcium modification. All of the modified alloys as well as the unmodified one perform the mechanism of complex of brittle and ductile fracture.

© 2018Lithium-sulfur (Li-S) cells have received particular attention as a “post lithium ion” energy storage system. However, low sulfur utilization and poor redox kinetics are still key challenges to improving cycling efficiency. Herein, we develop a multi-functional polysulfide mediator based on carbon hollow nanospheres supported titanium nitride (C@TiN) dual-shell hollow nanospheres, in which the physical confinement, chemical adsorption, and catalysis for sulfur species conversion were successfully achieved simultaneously. As a result, C@TiN-S composites (approximately 70 wt% sulfur content) exhibit faster reaction kinetics and a higher polysulfide trap capability than that of C@TiO2-S composites when used as cathode materials in Li-S batteries. The C@TiN-S electrode delivers a reversible capacity of 453 mA h g−1, coupled with a high average Coulombic Efficiency (~ 99.0%). There is also limited capacitance decay (only 0.0033% per cycle), at a current density of 3 C, over 300 cycles. In particular, when the sulfur loading is increased to 4.2 mg cm−2, the C@TiN-S electrode can provide a high capacity of 820 mA h g−1 over 150 cycles at 0.2 C. DFT calculations reveal that the long-chain Li2S8 tend to break down into two shorter chain segments due to the strong interaction of TiN and LiPSs. Electrochemical analysis techniques indicate that TiN can effectively catalyze the reduction of polysulfide and the oxidation of Li2S during discharge and charge processes, respectively. Our work offers a new strategy to develop high-performance Li-S batteries based on multi-functional mediators.

© 2018 Background: Vascular complications are major causes of disability and death in people with diabetes mellitus (DM). Nitric oxide (NO) supplement may help prevent vascular complications and is an attractive treatment option for DM. Hydroxytyrosol (HT) is a major polyphenol in olive oil. It is mainly used as a dietary supplement because of its antioxidant effect. Purpose: We aimed to determine the effects of hydroxytyrosol nitric oxide (HT-NO) on oxidative stress and NO level as well as related mechanisms. Study Design/Methods: The effects of HT-NO on oxidative stress and NO level were examined by using diabetic mouse model and HUVECs. Results: Our results showed that HT-NO has antioxidant and NO-releasing activities in vitro and in DM mice. HT-NO not only decreased blood glucose and oxidative stress but also increased NO level and deacetylase Sirtuin 1 (SIRT1) expression in DM mice and high glucose (HG)-stimulated HUVECs. Further studies found that SIRT1 activation augmented the effect of HT-NO on eNOS phosphorylation in HG-stimulated HUVECs. However, the promotive effect of HT-NO on eNOS phosphorylation was abolished by SIRT1 knockdown. Most importantly, HT-NO inhibited reactive oxygen species (ROS) production through SIRT1 in HUVECs. The ROS scavenger enhanced the effect of HT-NO on eNOS phosphorylation. Conclusion: These results suggest that HT-NO regulates oxidative stress and NO production partly through SIRT1 in DM mice and HG-stimulated HUVECs.

© 2018 Elsevier GmbH Background: Allergic asthma and atherosclerosis represent different directions of inflammatory responses of CD4+ T cells, and allergic asthma accelerates atherosclerosis formation. Curcumin could ameliorate the progression of both atherosclerosis and allergic asthma. Purpose: We aimed to investigate the roles of curcumin in asthma-accelerated atherosclerosis plaque formation, and the change of CD4+ T-cell subsets in this process. Methods: Six to eight-week-old apolipoprotein E−/− (apoE−/−) mice were sensitized and challenged by ovalbumin (OVA) to establish an allergic asthma model, and then received curcumin or vehicle treatment for 8 weeks. Results: The accelerated atherosclerosis was induced by allergic asthma accompanied by increased T helper cell (Th)2 and Th17 cells and decreased regulatory T cells (Tregs) in the spleen. After the 8-week treatment with curcumin, the lesion areas in the aortic root in asthmatic mice significantly improved, and the elevated Th2 and Th17 cells significantly decreased, but Tregs markedly increased. Although curcumin treatment markedly reduced the interleukin (IL)-4 and IL-13 in serum and spleen, the elevated IL-17A did not decrease. Moreover, Th1 cells showed no significant change between different groups. The mRNA expression levels of M1 macrophage-related inflammatory factors IL-6, iNOS and IL-1β were markedly elevated in the spleens of asthmatic mice, but significantly decreased after the 8-week treatment with curcumin. Conclusion: Curcumin ameliorated the aggravation of atherosclerotic lesions and stabilised plaque by modulating the balance of Th2/Tregs in asthmatic apoE−/− mice.

On the pricing of the persistence of earnings components in China Scopus

© 2018 Elsevier B.V. We examine the market pricing of the persistence of earnings components in the Chinese stock market. We find that the market on average misprices the persistence of total earnings. By decomposing total earnings into components, we find that while the market shows no evident mispricing of the persistence of the industry-wide earnings component, it does misprice the persistence of firm-specific earnings, in particular, firm-specific cash flows. Moreover, the mispricing of firm-specific earnings is more pronounced in the sample of firms with low firm-level disclosure quality. Our findings indicate that “investor fixation” is not a primary driver of the mispricing of the persistence of earnings components in China; instead, this mispricing is largely driven by the disclosure quality difference between earnings components.

Numerical investigation on the application of elliptical tubes in a spiral-wound heat exchanger used in LNG plant Scopus

© 2018 Elsevier Ltd The elliptical tubes were proposed to use in a spiral-wound heat exchanger to boost the shell-side transfer rate with the decrease of shell-side pressure drop. The effects of major axis and minor axis of elliptical tubes on thermal hydraulic performance are numerically investigated. The results show that, compared with the circular tube, the SWHE with elliptical tubes have a better heat transfer ability when the shell-side pressure drop is equal to that with circular tubes. With the increase of the major axis of the elliptical tube, the friction factor f decrease firstly and then increase. While with the increase of the minor axis, the variations of the f have an opposite trend with the increase of the major axis. For the heat transfer, the Nu decreases firstly with the increase of major axis and then increases. However, Nu number increases monotonously with the minor axis because elliptical tube bundle promotes the shell-side turbulent mixing. Based on the numerical data, the multi-objective optimization is performed by genetic algorithm. The optimal results show that, by contrast with the original structure, the comprehensive performance (K/△P) of the optimized structures increases by an average of 315.4% and the pumping consumption decreases by an average of 65.5%.

Related biomarkers of neurocognitive impairment in children with obstructive sleep apnea Scopus

© 2018 Objectives: Different experiment approaches have demonstrated that children with obstructive sleep apnea (OSA) exhibit neurocognitive and behavioral deficits. This review summarized the potential biomarkers of OSA-associated neurocognitive impairment in children. Methods: A scoping review of studies on children with OSA that evaluated the potential value of different markers in identifying neurocognitive impairment was undertaken. Additionally, the biomarkers were categorized according to the different research methods, including brain imaging studies, serological indicators and urine markers. Results: Majority of the studies that evaluated blood biomarkers, plasma insulin growth factor-1 (IGF-1) and Alzheimer's disease (AD)-related biomarkers appeared to exhibit a favorable profile, and could discriminate between OSA children with or without neurocognitive impairments. Brain imaging studies and urinary neurotransmitters could also be helpful for screening OSA cognitive morbidity in children. Conclusion: Due to limited research methods available in children, the cognitive susceptibility of children with OSA has been rarely studied. The main reason for this may be the limited research methods in children. Numerous study populations of children and complex psychological tests are required, which involve major labor and costs.Multi-center prospective studies are needed to identify suitable biomarkers for the timely prediction and effective intervention to prevent neurocognitive impairment in children with OSA and to explore further opportunities in this arena.

© 2018 Elsevier Ltd In this experimental study, the after-treatment characteristics of urea-based selective catalytic reduction (urea-SCR) in a diesel engine was investigated at a low engine exhaust temperature (250 °C). This represented the idle state of diesel vehicles during road congestion. Detailed experimental work was performed on a heavy-duty diesel engine test bench under the World Harmonized Transient Cycle. The influence of the ammonia/nitrogen ratio was first investigated using vanadium-based SCR (V-SCR) catalyst. When the ammonia/nitrogen ratio was 1.0, the influences of space velocity on the after-treatment characteristics of two different configurations of V-SCR catalysts were investigated in terms of NOX emission, NH3 slip and NOX conversion efficiency. Small SV resulted in reduction of the NH3 slip and the coated-type catalyst indicated more NH3 slip than the extruded-type catalyst at equal catalyst dimensions. It was concluded that the extruded-type V-SCR catalyst showed better operating characteristics than did the coated-type V-SCR catalyst.

© 2019 Institute for Scientific Computing and Information. A posteriori error estimators are studied for discontinuous Galerkin methods for solving a representative elliptic variational inequality of the second kind, known as a simplified frictional contact problem. The estimators are derived by relating the error of the variational inequality to that of a linear problem. Reliability and efficiency of the estimators are theoretically proved.
