Saikosaponin A inhibits compound 48/80-induced pseudo-allergy via the Mrgprx2 pathway in vitro and in vivo SCIE PubMed Scopus

Abstract :

Pseudo-allergic reactions-adverse, non-immunologic, anaphylaxis-like sudden onset reactions mediated through an IgE-independent pathway are activated by various basic compounds and occur at least as frequently as IgE-mediated reactions to drugs. A large family of G protein coupled receptors (Mas-related genes: Mrgprs) is closely related to pseudo-allergies. However, few therapies can directly target pseudo-allergies and related Mrgprs. Saikosaponin A (SSA) is effective in the treatment of passive cutaneous anaphylaxis (PCA), adjuvant arthritis, and delayed hypersensitiveness. In this study, we investigated the anti-pseudo-allergy effect of SSA and its underlying mechanism. We examined the effect of SSA on both IgE-independent and IgE-dependent responses using PCA and active systemic anaphylaxis models, as well as in vitro-cultured mast cells. We also evaluated whether the anti-allergy effect is related to Mrgprs by using in vitro Mrgprx2-expressing HEK293 cells. SSA dose dependently suppressed compound 48/80 (C48/80)-induced PCA and mast cell degranulation in mice. When SSA and C48/80 were administered together through the vein, C48/80-induced systemic anaphylaxis did not occur, and C48/80-induced shock ratio decreased dose-dependently upon SSA treatment. However, SSA did not affect IgE-dependent allergy. When administered topically 24 h before antigen challenge, Evans blue leakage and paw swelling were induced in the SSA-treated group and the vehicle group. Our in vitro studies revealed that SSA reduced C48/80-induced calcium flux and suppressed degranulation in LAD2 cells. SSA could also dose-dependently inhibit C48/80-induced Mrgprx2-expressing HEK293 cell activation. As a conclusion, SSA could inhibits IgE-independent allergy, but not IgE-dependent allergy, and this effect involves the Mrgprx2 pathway. Abstract :

Traditional Chinese medicine (TCM) has been used for prevention and treatment of various diseases for many decades. TCM injection is a new dosage form, with incidence of anaphylactoid reactions increasing every year. In this study, the rat basophilic leukemia 2H3 (RBL-2H3) and laboratory of allergic disease 2 (LAD2) dual-mixed/CMC was established and was coupled with an HPLC-ESI-IT-TOF-MS system to identify the potential allergenic components in Haqing injection. Cinobufagin, piperine, osthole, praeruptorin A, and schizandrin A were screened from Haqing injection via this coupled system. Competitive binding assay showed piperine, praeruptorin A, and schizandrin A acting on MrgprX2 and cinobufagin and osthole act on the IgE receptor. The release of mediators of anaphylaxis results showed cinobufagin and osthole can cause anaphylactoid reactions by triggering the release of -hexosaminidase and histamine via IgE-R. Praeruptorin A and schizandrin A could promote the release of -hexosaminidase and histamine via MrgprX2 receptor. In summary, the dual-mixed/CMC model can significantly improve the efficiency of target component identification from a complex sample. When combined with competitive binding assay and validation of biological activities, this model enables accurate determination of the dual-target components, offering improved methods for quality control of TCM injections.

Salvianolic acid A (Sal A) has been shown to prevent and treat ischemic cardiovascular, as well as cerebral vascular diseases. However, little is known about Sal A in renal ischemia/reperfusion (I/R) injury. In this study, a renal I/R injury model in rats and a hypoxia/reoxygenation (H/R) model to damage proximal renal tubular cells (HK-2) were used to assess whether Sal A halts the development and progression of renal I/R injury. As compared with vehicle treatment, Sal A significantly attenuated kidney injury after renal I/R injury, accompanied by decreases in plasma creatinine, blood urea nitrogen levels, the number of apoptosis-positive tubular cells, and kidney oxidative stress. Sal A also activated phosphorylated protein kinase B (p-Akt) and phosphorylated-mammalian target of rapamycin (p-mTOR) compared with vehicle-treated I/R injury rats. In H/R-injured HK-2 cells. Sal A can reduce the levels of reactive oxygen species in a dose-related manner. Similar to the results from in vivo experiments, in vitro Sal A also increased the protein expression of phosphorylated-eukaryotic initiation factor 4E binding protein 1 (p-4EBP1) compared with vehicle. Furthermore. the cytoprotective activity of Sal A was inhibited by LY294002 and rapamycin. These findings indicate that Sal A can ameliorate renal I/R injury and promote tubular cell survival partly via the Akt/mTOR/4EBP1pathway. Sal A could be a candidate compound to prevent ischemic tissue damage.

Abstract :

Adverse drug reactions of Danshen injection mainly manifested as pseudoallergic reactions. In the present study, salvianolic acid A and a pair of geometric isomers (isosalvianolic acid C and salvianolic acid C) were identified as pseudoallergic components in Danshen injection by a high-expression Mas-related G protein coupled receptor X2 cell membrane chromatography coupled online with high-performance liquid chromatography with electrospray ionization tandem mass spectrometry. Their pseudoallergic activities were evaluated by in vitro assay, which were consistent with the retention times on the cellmembrane chromatography column. Salvianolic acid C, the most outstanding compound, was further found to induce pseudoallergic reaction through Mas-related G protein coupled receptor X2. All the results above indicated that the system developed in this study is an effective method for simultaneously analyzing pseudoallergic components, even those with similar structures and the microcomponents in complex samples (salvianolic acid C in Danshen injection).

Rhizoma Belamcandae (Shegan), obtained from the dried rhizome of Belamcanda chinensis (L.) DC., is one of the most important traditional Chinese medicines (TCMs). It has many pharmacological effects, such as eliminating toxins, heat, phlegmand dyspnea, antiviral effect, anti-inflammatory, etc. In recent years, studies on Shegan were increasing, and reports showed that tectorigenin and tectoridin separated from Shegan had the activity of inhibiting angiogenesis, however, the targets which they act with were uncertain. Hence, screening receptor-specific components with anti-tumor effect from Shegan is necessary. Epidermal growth factor receptor (EGFR) is a part of the tyrosine kinase family of receptors responsible for intracellular signalling and cell growth, and overexpression of EGFR is associated with angiogenesis, invasion and metastasis of cancer cells. Cell membrane chromatography (CMC) has its history of more than 20 years, it was established in 1996 by professor He and his colleagues. As a kind of biological affinity chromatography, CMC has been developed and confirmed to be an effective method for screening bioactive components from complex systems. In this study, a two-dimensional online system was built to screen active compounds acting on epidermal growth factor receptor from Rhizoma Belamcandae. The first dimension was EGFR/CMC system, and the second was HPLC-IT-TOF MS system. Gefitinib was selected as the positive control drug to investigate the suitability of the EGFR/CMC-online-HPLC-IT-TOF MS system. For the EGFR/CMC system, the EGFR HEK293 engineered cell line was cultured and used to prepare the EGFR/CMC column according to the wet packing method, after analysing total extracts of Shegan through the EGFR/CMC system, the retained fractions were switched to the second dimension for identification. Molecular docking assay was performed using the SYBYL-X1.1software to determine the strength of binding between the screened components and EGFR, and methyl thiazolyl tetrazolium (MTT) assay were used to evaluate the in vitro inhibition activity of the target components. The screening results showed that irisflorentin from Rhizoma Belamcandae was the targeted component which could specifically act on EGFR. Molecular docking assay showed irisflorentin could act on EGFR in similar manner with gefitinib as a positive control drug. The results of MTT assay showed gefitinib and irisflorentin inhibited high expressed EGFR cell growth in a dose-dependent manner from 0.4 to 50.0 μmol/L. These results indicated that EGFR cell membrane chromatography online high performance liquid chromatography-ion trap-time of flight-mass spectrometry (EGFR/CMC-online-HPLC-IT-TOF MS) system would be a useful method in drug discovery with natural medicinal herbs for searching potential anti-tumor candidates. © 2017, Editorial Board of Journal of Chinese Mass Spectrometry Society. The equilibrium dissociation constant (ICD) of drug-membrane receptor affinity is the basic parameter that reflects the strength of interaction. The cell membrane chromatography (CMC) method is an effective technique to study the characteristics of drug-membrane receptor affinity. In this study, the KD value of CMC relative standard method for the determination of drug-membrane receptor affinity was established to analyze the relative ICD values of drugs binding to the membrane receptors (Epidermal growth factor receptor and angiotensin II receptor). The K-D values obtained by the CMC relative standard method had a strong correlation with those obtained by the frontal analysis method. Additionally, the ICp values obtained by CMC relative standard method correlated with pharmacological activity of the drug being evaluated. The CMC relative standard method is a convenient and effective method to evaluate drug membrane receptor affinity. (C) 2017 Elsevier B.V. All rights reserved.

Mast cells are unique immune cells that function as sentinels in host defence reactions, including immediate hypersensitivity responses and allergic responses. The mast cell-specific receptor named MAS-related G protein-coupled receptor X2 (MRGPRX2) triggers mast-cell degranulation, a key process in anaphylactoid reactions. It is widely observed that antimicrobials can induce pseudo-allergic reactions (i.e. IgE-independent mechanism) with symptoms ranging from skin inflammation to life-threatening systemic anaphylaxis. However, their direct involvement and the mechanisms underlying anaphylactoid reactions caused by antimicrobials have not been demonstrated. Structurally different antimicrobials were screened by Ca2+ imaging using MRGPRX2 overexpressing HEK293 cells. MRGPRX2 related anaphylactoid reactions induced by these components were investigated by body temperature drop and mast cell degranulation assays. We showed that MRGPRX2 is involved in allergic-like reactions to three types of antimicrobials in a dose-dependent manner. However, mast cells lacking the receptor show reduced degranulation. Furthermore, mice without MAS-related G protein-coupled receptor B2 (the orthologous gene of MRGPRX2) exhibited reduced substance-induced inflammation. Interestingly, beta-lactam and antiviral nucleoside analogues did not induce anaphylactic reactions, which were also observed in vitro. These results should alarm many clinicians that such drugs might induce anaphylactoid reactions and provide guidance on safe dosage of these drugs.

MrgprB2 knockout mice exhibit a reduced SH-induced inflammation effect. Furthermore, MRGPRX2 (the orthologous gene of MrgprB2) related human mast cells are activated by SH in a dose-dependent manner; however, MRGPRX2 knockdown mast cells showed reduced degranulation. The results showed a kind of mechanism that SH-induced anaphylactoid reactions were mediated by MRGPRX2 via activating PLC molecular signaling pathways to provoke mast cells Ca2+ mobilization and degranulation. Anaphylactoid reactions are common clinical acute adverse drug reactions that can exacerbate a patient's condition and produce effects that may become life-threatening. Therefore, it is important to establish a novel method to evaluate drugs for anaphylactoid reactions. In this study, we developed a sensitive and rapid method to detect histamine release from LAD2 cells using liquid chromatography-tandem mass spectrometry (LC-MS/MS) and constructed a relative release index based on various release curve parameters, including allergen release time and sudden change rate, to evaluate the potential and strength of allergen-induced anaphylactoid reactions. This LAD2 release model was used to evaluate anaphylactoid reactions induced by ciprofloxacin, norfloxacin, lomefloxacin, moxifloxacin, and baicalin. The results positively correlated with those obtained with an Evans blue ear test and negatively correlated with the Ca2+ influx EC50. In summary, the current study established a novel in vitro method to analyze the properties of histamine release from LAD2 cells and characterize the sensitization and strength of sensitization of drugs or components that may induce anaphylactoid reactions.

Abstract :

A new approach for the surficial-modification of silica with 5-HMF as a functional molecule was first designed. The infrared spectrum, X-ray photoelectron spectroscopy and elemental analysis showed that the modified-SiO2 particles were successfully established. Full dispersion of SiO2 and the introduction of gallic acid were confirmed to be favorable for the 5-HMF content. The retention behavior of bovine serum albumin on the modified-SiO2 particle suggested that the novel particles could serve as a promising stationary phase on the identification of the particular proteins.
